Villain in this hand just doesn't seem that good. His flop bet is horrendously small (is this a normal C-bet by him or does he bet small when he flops a big hand?). His turn check is suspect, but not terrible on occasion. I like the check/raise by him on the river because he must be like 80% certain someone has a flush, and his line looks a bit FOS and will get looked up a lot.
Anyway, like Zeestein says, this is almost never a bluff unless you are playing a very strong player that knows you are able to make HUGE laydowns. Villain should know your hand is a boat here about 0% of the time.
