The 100 buyins are quite a bit better than the 25s, but you can establish similar winrates by stealing small pots and picking up the occasional big one. You should be able to beat them easy enough, but your table selection needs to be good - I wouldn't look at any with an AP less than $30.
I'm not sure what relevance winnings has to your plans, whats your BR? And a loss of 2.5 buyins gives you too little wiggle room, I think. You could easily lose that in a bad hour. Plan to maybe put $1k aside for the 100 games, and shift back down if you lose that. Is that possible? If your BR is restricted, perhaps you could choose to play with maybe 4 buyins in a "taking a stab" sort of way; if you double that through, move up permanently, if not, build up again at PLO25 then move back.
Why are you leapfrogging PLO 50? Jumping two levels IMO could leave you playing scared and not at your best, at least if you're anything like me.
Interested to here how you get on! You are a winning 25 player and i'm sure you can beat the 100 games with good game selection, would be nice to hear more details about your current financial situation and aims.
